چکیده

Multidrug resistance can result from several factors and processes. Some types of multidrug resistance (MDR) were recognized whose patterns differ from pattern of P-glycoprotein (Pgp) associated MDR in several important ways. Although, there is limited evidence for effect of DHP on atypical MDR; based on some observations like noncompetitive inhibition of topoisomerase I by a DHP derivative (Dexniguldipine), it has been hypothesized that some DHP derivatives which have essential structural feature have possible effect on altering the topoisomerase II levels or activity which results in reversing topoisomerase–mediated MDR. If this hypothesis is verified, some new dihydropyridine can be synthesized with possible dual effect on both typical and atypical MDR.

CRISPR/Cas9, a new approach to successful knockdown of ABCB1/P-glycoprotein and reversal of chemosensitivity in human epithelial ovarian cancer cell line

Objective(s): Multidrug resistance (MDR) is a major obstacle in the successful chemotherapy of ovarian cancer. Inhibition of P-glycoprotein (P-gp), a member of ATP-binding cassette (ABC) transporters, is a well-known strategy to overcome MDR in cancer. The aim of this study was to investigate the efficiency and ability of CRISPR/Cas9 genome editing technology to knockdown ABCB1 gene expression ...

Specific reversal of MDR1/P-gp-dependent multidrug resistance by RNA interference in colon cancer cells.

No control cell line was available for previous RNA interference studies on reversal of multidrug resistance (MDR) in colon cancer cells. Here, human COLO 320DM, with HT-29 as the control, colon cancer cell lines were used to investigate the reversal of MDR1/P-gp-dependent MDR by siRNA (#4123 and #4029 MDR1 siRNAs) targeting to MDR1 mRNA. Both siRNAs inhibited expression of MDR1 and P-gp in COL...

Quantitative structure-activity relationship of multidrug resistance reversal agents.

Multidrug resistance (MDR) is one of the major obstacles to long term successful cancer chemotherapy. The use of MDR reversal (MDRR) agents is a promising approach to overcome the undesired MDR phenotype. To design more effective MDRR agents that are urgently needed for clinical use, a data set of 609 diverse compounds tested for MDRR activity against P388/ADR-resistant cell lines was submitted...

Overcoming PGP-related multidrug resistance. The cyclosporine derivative SDZ PSC 833 can abolish the resistance to methoxy-morpholynil-doxorubicin.

BACKGROUND The results obtained so far in studies designed to neutralize P glycoprotein (PGP)-related multidrug resistance (MDR) by using MDR reversal agents, have not yet fulfilled the promise of the experiments which were performed in vitro. In order to improve PGP-related MDR neutralization, we tested in vitro the activity of the cyclosporine derivative SDZ PSC 833 (PSC) together with doxoru...

Targeting the ABCG2-overexpressing multidrug resistant (MDR) cancer cells by PPARγ agonists.

BACKGROUND AND PURPOSE Multidrug resistance (MDR), usually mediated by overexpression of efflux transporters such as P-gp, ABCG2 and/or MRP1, remains a major obstacle hindering successful cancer chemotherapy. There has been great interest in the development of inhibitors towards these transporters to circumvent resistance. However, since the inhibition of transporter is not specific to cancer c...
